What is Kisspeptin?
Kisspeptin refers to a group of neuropeptides — most commonly studied as Kisspeptin-10 and Kisspeptin-54 — encoded by the KISS1 gene. They act on the GPR54 receptor (also known as KISS1R) on GnRH neurons in the hypothalamus.
Kisspeptin sits at the very top of the hypothalamic-pituitary-gonadal (HPG) axis, acting as the master gatekeeper of GnRH release.
Mechanism of action
Binding of kisspeptin to GPR54 triggers GnRH secretion from hypothalamic neurons. GnRH then stimulates pulsatile LH (and FSH) release from the anterior pituitary, which in turn drives gonadal testosterone or estradiol production.
Because kisspeptin acts upstream of GnRH, it has been studied as a potential tool for probing HPG axis function in hypogonadism, hypothalamic amenorrhea, and assisted reproduction research.
Half-life
Kisspeptin-10 has a plasma half-life of roughly 4 minutes. Kisspeptin-54, the full-length endogenous form, has a longer half-life of approximately 25–30 minutes and is more commonly used in clinical research studies for that reason.
